Use of exosomes (extracellular vesicles) to specifically target Her2+ve breast cancer for gene (mRNA) delivery. The mRNA encodes an improved novel nitroreductase and converts our newly discovered highly effective prodrug, CNOB, into the cytotoxic MCHB. The latter can be imaged in live mice, thereby greatly facilitating detection of off-target delivery of the mRNA and permitting rapid screening of different approaches to achieve specificity. The project involves several Stanford cancer researchers and is part of a nationwide NIH consortium.
